The Protective Effect of Silymarin on Lipopolysaccharide-Induced Liver Toxicity in Male Wistar Rat
Authors
Abstract:
Background and Aims: Lipopolysaccharide (LPS) triggers production of reactive oxygen species and inflammatory cytokines. Nowadays Silybum marianum has been shown to treat liver and gall bladder disorders, especially to protect the liver against poisoning from various toxins. Therefore, we decided to evaluate the protective effect of silymarin on LPS-induced liver toxicity in male Wistar rat. Materials and Methods: Totally, 40 male Wistar rats were divided into 4 groups (n=10 in each): The animals were treated with silymarin for two weeks before the biochemical tests. Apoptosis was assessed by evaluating the amount of proteins in liver tissues by western blotting. Results: LPS induced hepatotoxicity as evidenced by histopathological damages and biochemical abnormalities. Data showed that malondialdehyde level significantly increases in the liver of LPS-treated rats. Destructive effects of LPS on histopathological and biochemical parameters were improved. LPS also increased expression of Bax/Bcl2 ratio and activation of caspase 3, caspase 8 and caspase 9. Western blot analysis showed silymarin treatment inhibiting apoptosis stimulated by LPS in the liver (p<0.001). Conclusions: The results of this research demonstrated that silymarin can exert protective effects against toxic effects of LPS in rat liver. Anti-inflammatory drug can play a protective role in attenuating the liver inflammation induced by LPS injection.

The present investigation aims at assessing the hepatoprotective effect of triterpene fraction isolated from the stem bark of Wrightia tinctoria (containing lupeol, b -amyrin and b -sitosterol) on CCl 4 -induced hepatotoxicity in the rat. CCl 4 (1.5 mg/kg, i.p) is a potent hepatotoxic agent which causes peroxidative degeneration of membrane lipids with the potential outcome of fatt...
full textThe Protective Roles of Zinc and Magnesium in Cadmium-Induced Renal Toxicity in Male Wistar Rats
Background: Cadmium (Cd) is a heavy metal that has widespread use. It enters the food chain in different ways, including soil and water. Cadmium can cause dysfunction of different body organs. Zinc (Zn) and magnesium (Mg) supplementation can have protective effects against cadmium toxicity due to their antagonistic and antioxidants properties. This study examines the influence of supplemental...
full textPreventive effect of novel nanomissile of silymarin on liver injury induced by carbon tetrachloride in rat
Background and Aim: Silymarin, obtained from Silybum marianum has hepatoprotective effects and antioxidant activity and has been used in the treatment of liver diseases. Because of its poor aqueous solubility and low bioavailability its use has been limited. The aim of this study was to prepare a new and inexpensive formulation of silymarin using simple dissolution methods and compare its hepat...

Objective: Neuroimmune factors contribute on the pathogenesis of sickness behaviors. Nigella sativa (NS) has anti-inflammatory, anti-anxiety and anti-depressive effects. In the present study, the effect of NS hydro-alcoholic extract on sickness behavior induced by lipopolysaccharide (LPS) was investigated. Materials and Methods: The rats were divided into five groups (n=10 in each): (1) control...
full textAntihyperlipidemic Effect of Wheatgrass on Alcohol and ∆pufa Induced Liver Toxicity in Male Albino Wistar Rats
Objective: Alcohol is one of the widely used and abused drugs in the society. Chronic alcohol consumption causes various hepatic lesions that are grouped under common term, alcoholic liver diseases. Dietary intake of fats, especially repeatedly heated polyunsaturated fatty acid (∆PUFA) produces various lipid peroxidative end products which can aggravate the pathological changes produced by etha...
